Johann Baptist von Spix (H\u00f6chstadt an der Aisch, 9 de fevereiro 1781 \u2014 14 de mar\u00e7o de 1826) foi um naturalista alem\u00e3o.&lt;\/p&gt;\n&lt;p&gt;Em 1817, Spix e o seu colega Carl von Martius foram convidados para realizar uma expedi\u00e7\u00e3o ao Brasil, com o objectivo de descrever sua fauna e flora. Compunham a Miss\u00e3o Art\u00edstica Austro-Alem\u00e3 que acompanhou a princesa Maria Leopoldina de \u00c1ustria, futura imperatriz do Brasil. A viagem terminou em 1820 e Spix voltou para a Europa com cerca de 9 000 esp\u00e9cies de plantas e animais, incluindo mam\u00edferos, aves e anf\u00edbios. O conjunto foi a base da colec\u00e7\u00e3o do Museu de Hist\u00f3ria Natural de Munique. Entre 1817 e 1820, o bot\u00e2nico Carl Friedrich Philipp von Martius (1794-1868) e o zo\u00f3logo Spix, ambos origin\u00e1rios do estado germ\u00e2nico da Baviera, participaram da chamada expedi\u00e7\u00e3o austr\u00edaca pelo interior do Brasil, promovida em raz\u00e3o da uni\u00e3o entre a arquiduquesa Maria Leopoldina e o pr\u00edncipe herdeiro e futuro imperador do Brasil, dom Pedro.<\/span><\/p>\n<h3><span style=\"color: #000000;\"><strong><span style=\"font-family: 'courier new', courier, monospace; font-size: 12pt;\">Um mico como presente<\/span><\/strong><\/span><\/h3>\n<p><span style=\"color: #000000;\">A viagem come\u00e7ou no Rio de Janeiro, ent\u00e3o capital nacional, e percorreu partes do Sudeste, do Nordeste e do Norte. Perto do final de suas andan\u00e7as pela grande floresta tropical, \u00faltima etapa de seu p\u00e9riplo, a dupla de naturalistas separou-se depois de ter chegado \u00e0 localidade de Tef\u00e9, no Amazonas. Martius subiu o rio Japur\u00e1, mais ao norte, antes de retornar para Manaus, em mar\u00e7o de 1820. M\u00fcller<\/span><\/figcaption><\/figure>\n<p><span style=\"color: #000000;\">Nesse ponto da floresta, perto da fronteira com a Col\u00f4mbia, o zo\u00f3logo recebeu como presente, provavelmente dos ind\u00edgenas Tikuna, um exemplar de um diminuto macaco, morto. Com cerca de 15 cent\u00edmetros de comprimento (exclu\u00eddo o rabo) e por volta de 100 gramas de peso, o primata foi descrito por Spix e recebeu o nome de\u00a0<em>Cebuella pygmaea<\/em>. Em seu trabalho, o naturalista indicou que o animal era proveniente da regi\u00e3o de Tabatinga. Por\u00e9m n\u00e3o deixou claro se o sagui viera da margem direita ou esquerda do Solim\u00f5es.<\/span><\/p>\n<p><span style=\"color: #000000;\">Nos anos 1940, o zo\u00f3logo sueco Einar L\u00f6nnberg (1865-1942) prop\u00f4s a exist\u00eancia de outra esp\u00e9cie muito similar \u00e0 descrita por Spix: um sagui-pigmeu de ventre branco, cujo exemplar estudado pelo pesquisador escandinavo era origin\u00e1rio da localidade de Eirunep\u00e9, distante 200 quil\u00f4metros ao sul de Tabatinga, portanto proveniente da margem direita (sul) do rio Solim\u00f5es. Os dois tipos de pequenos primatas divergiram, em termos evolutivos, h\u00e1 2,5 milh\u00f5es de anos. \u201cPassamos a considerar que havia duas esp\u00e9cies distintas de sagui-pigmeu\u201d, diz Boubli. Mas n\u00e3o estava claro se o sagui descrito por Spix deveria permanecer com o nome de\u00a0<em>C. pygmaea<\/em>\u00a0ou adotar o de\u00a0<em>C. niveiventris<\/em>. Essa quest\u00e3o foi resolvida pelo novo estudo do zo\u00f3logo brasileiro.<\/span><\/p>\n<p><span style=\"color: #000000;\">Al\u00e9m de analisar o DNA do esp\u00e9cime coletado por Spix, o artigo da\u00a0<em>Zoological Research\u00a0<\/em>comparou-o ao material gen\u00e9tico de 65 exemplares atuais de macacos das Am\u00e9ricas, entre os quais 52 do g\u00eanero\u00a0<em>Cebuella<\/em>. O estudo indicou que os exemplares de sagui-pigmeu com barriga amarela eram todos provenientes de \u00e1reas ao norte do Solim\u00f5es e os com ventre branco tinham seu h\u00e1bitat ao sul desse rio. Com esse dado em m\u00e3os, bastaria, ent\u00e3o, olhar para a tonalidade do ventre do exemplar dado a Spix, mantido na Cole\u00e7\u00e3o de Zoologia do Estado da Baviera, em Munique, para determinar sua esp\u00e9cie.<\/span><\/p>\n<h4><strong><span style=\"color: #000000; font-family: 'courier new', courier, monospace; font-size: 12pt;\">Esp\u00e9cime-tipo de import\u00e2ncia hist\u00f3rica<\/span><\/strong><\/h4>\n<p><span style=\"color: #000000;\">\u201cMas apenas examinar o exemplar n\u00e3o resolvia a quest\u00e3o porque n\u00e3o dava mais para ver a colora\u00e7\u00e3o da barriga. Se tiv\u00e9ssemos certeza de que Spix coletou ou ganhou o sagui na margem esquerda do rio, tamb\u00e9m poder\u00edamos resolver a taxonomia sem problemas. No entanto, no in\u00edcio do s\u00e9culo XIX, n\u00e3o se dava muita aten\u00e7\u00e3o para a localidade das amostras. Sempre se assumiu que o sagui de Spix era de Tabatinga, mas isso nunca esteve registrado em lugar nenhum. Essa dificuldade foi contornada com a entrada em cena do geneticista Christian Roos, do Centro de Primatas da Alemanha. Especialista em an\u00e1lise de DNA antigo e com tr\u00e2nsito em Munique, Roos conseguiu uma autoriza\u00e7\u00e3o dos respons\u00e1veis pela cole\u00e7\u00e3o b\u00e1vara de zoologia para retirar um pedacinho de tecido do pequeno sagui.<\/span><\/p>\n<h4><strong><span style=\"color: #000000; font-family: 'courier new', courier, monospace; font-size: 12pt;\">DNA mitocondrial<\/span><\/strong><\/h4>\n<p><span style=\"color: #000000;\">A amostra forneceu material suficiente para extrair seu DNA mitocondrial, que tem cerca de 12 mil pares de bases. Essa parte do genoma \u00e9 herdada apenas da linhagem materna e, em termos evolutivos, \u00e9 bastante conservada ao longo das gera\u00e7\u00f5es, particularidade que a torna uma ferramenta molecular \u00fatil para entender as rela\u00e7\u00f5es entre diferentes esp\u00e9cies. O DNA mitocondrial do sagui coletado por Spix h\u00e1 200 anos era praticamente id\u00eantico aos dos exemplares atuais de\u00a0<em>C. pygmaea<\/em>, constata\u00e7\u00e3o que encerrou a discuss\u00e3o taxon\u00f4mica a respeito de sua esp\u00e9cie.<\/span><\/p>\n<p><span style=\"color: #000000;\">Para a bioantrop\u00f3loga norte-americana Leila Porter, da Universidade do Norte de Illinois, nos Estados Unidos, que estuda a ecologia e o comportamento de primatas, a an\u00e1lise do DNA de exemplares mantidos em cole\u00e7\u00f5es biol\u00f3gicas deve se consolidar como um procedimento padr\u00e3o em estudos comparativos. <\/span><\/p>\n<blockquote><p><span style=\"color: #ffffff;\"><strong><span style=\"font-family: 'courier new', courier, monospace; font-size: 10pt;\">\u201cNo futuro, os primatologistas continuar\u00e3o a usar DNA antigo coletado de amostras de museus para resolver incertezas taxon\u00f4micas\u201d, disse Porter em entrevista a\u00a0<em>Pesquisa FAPESP<\/em>. A pesquisadora \u00e9 coautora do artigo rec\u00e9m-publicado sobre os saguis-pigmeus da Amaz\u00f4nia. \u201cSe poss\u00edvel, eles tamb\u00e9m dever\u00e3o obter material gen\u00e9tico de amostras fecais das popula\u00e7\u00f5es de animais dos locais de estudo. A exemplo do mist\u00e9rio envolvendo os saguis-pigmeus do oeste da Amaz\u00f4nia brasileira, existem v\u00e1rios outros enigmas que tamb\u00e9m poder\u00e3o ser desvendados, ou, ao menos, mais bem compreendidos, por meio de estudos com material gen\u00e9tico mais antigo. \u201cQueremos obter DNA de esp\u00e9cimes-tipos de macacos brasileiros que est\u00e3o em museus sem uma identifica\u00e7\u00e3o geogr\u00e1fica clara. Em 1848, aos 25 anos, Wallace embarcou ao lado de seu compatriota Henry Walter Bates (1825-1892) do porto de Liverpool com destino ao Par\u00e1 para participar de uma expedi\u00e7\u00e3o pelo interior da Amaz\u00f4nia.<\/span><\/p>\n<p><span style=\"color: #000000;\">Seguindo as pistas dos grandes exploradores, Boubli pretende estudar se ocorre hibrida\u00e7\u00e3o natural entre as duas esp\u00e9cies de sagui-pigmeu nas cabeceiras dos rios da Amaz\u00f4nia. Em tese, duas esp\u00e9cies distintas devem ter perdido a capacidade de cruzar e gerar descendentes f\u00e9rteis. Mas esses limites biol\u00f3gicos nem sempre s\u00e3o t\u00e3o r\u00edgidos na natureza. H\u00e1 relatos de v\u00e1rios pesquisadores de que em algumas regi\u00f5es existem saguis com barriga de padr\u00e3o bicolor, uma mistura de amarelo e de branco.
