Embora muitos organismos sejam capazes de regenera\u00e7\u00e3o parcial, todos, sem exce\u00e7\u00e3o, enfrentam o processo inexor\u00e1vel do envelhecimento. Nas \u00faltimas d\u00e9cadas, a biologia celular e <a href=\"https:\/\/biologo.com.br\/bio\/biologia-molecular\/\"><span style=\"text-decoration: underline;\">molecular<\/span><\/a> revelou as engrenagens desse mecanismo silencioso, delimitando suas causas, consequ\u00eancias e, sobretudo, suas possibilidades de modula\u00e7\u00e3o.<\/p>\n\n\n\n<p class=\"has-black-color has-text-color has-link-color has-medium-font-size wp-elements-6e2f92a5cc49a51f60ef649103cb49c7\">O envelhecimento celular, tamb\u00e9m conhecido como senesc\u00eancia, \u00e9 um estado em que a c\u00e9lula perde a capacidade de se dividir, mesmo quando ainda est\u00e1 metabolicamente ativa. Inicialmente, essa resposta foi compreendida como um mecanismo de prote\u00e7\u00e3o contra a prolifera\u00e7\u00e3o descontrolada de c\u00e9lulas danificadas, como ocorre no c\u00e2ncer. A cada divis\u00e3o celular, os tel\u00f4meros encurtam at\u00e9 um ponto cr\u00edtico, quando a c\u00e9lula entra em senesc\u00eancia replicativa (Blackburn et al., 2015). Nesse contexto, a enzima telomerase, capaz de restaurar essas sequ\u00eancias, passou a ser vista como chave para a longevidade celular. No entanto, seu uso desregulado tamb\u00e9m se associa \u00e0 imortalidade de c\u00e9lulas tumorais, o que imp\u00f5e limites \u00e9ticos e biol\u00f3gicos \u00e0 sua aplica\u00e7\u00e3o terap\u00eautica.<\/p>\n\n\n\n<h3 class=\"wp-block-heading has-medium-font-size\"><strong>Mecanismos do envelhecimento<\/strong><\/h3>\n\n\n\n<p class=\"has-black-color has-text-color has-link-color has-medium-font-size wp-elements-464ca7aab512d3875344f59b97336b23\">Ao mesmo tempo, outros fatores contribuem para o envelhecimento do metabolismo. Por exemplo, o ac\u00famulo de esp\u00e9cies reativas de oxig\u00eanio (ROS) provoca danos oxidativos em DNA, prote\u00ednas e lip\u00eddios. Embora o organismo possua defesas antioxidantes, estas se tornam menos eficientes com o passar dos anos. Como resultado, os danos se acumulam e afetam o funcionamento celular global. Com o tempo, marcas qu\u00edmicas sobre o DNA, como a metila\u00e7\u00e3o, se desorganizam, afetando a express\u00e3o de genes essenciais para a manuten\u00e7\u00e3o da integridade celular. Consequentemente, a c\u00e9lula perde sua identidade funcional, o que contribui para a degenera\u00e7\u00e3o tecidual. Al\u00e9m disso, o mau funcionamento das mitoc\u00f4ndrias \u2014 respons\u00e1veis pela produ\u00e7\u00e3o de energia \u2014 intensifica o ciclo de degrada\u00e7\u00e3o.<\/p>\n\n\n\n<h3 class=\"wp-block-heading has-medium-font-size\"><strong>Senesc\u00eancia <\/strong><\/h3>\n\n\n\n<p class=\"has-black-color has-text-color has-link-color has-medium-font-size wp-elements-70b1b398c5baff5d17ed035ec9841e23\">Entretanto, a senesc\u00eancia n\u00e3o atua apenas como um marcador de deteriora\u00e7\u00e3o. Diversos estudos indicam que c\u00e9lulas senescentes secretam um conjunto de sinais inflamat\u00f3rios e remodeladores do ambiente, conhecido como SASP (<em>senescence-associated secretory phenotype<\/em>). Embora esse fen\u00f3tipo possa ser \u00fatil na cicatriza\u00e7\u00e3o e na vigil\u00e2ncia tumoral, ele tamb\u00e9m contribui para processos inflamat\u00f3rios cr\u00f4nicos, como a osteoartrite e a aterosclerose (Campisi, 2013).<\/p>\n\n\n\n<figure class=\"wp-block-pullquote has-white-color has-text-color has-link-color has-large-font-size wp-elements-6eac786d6c31ad809e9fb37957d2e8b2 has-background\" style=\"border-radius:86px;background-image:url(&apos;https:\/\/biologo.com.br\/bio\/wp-content\/uploads\/2018\/11\/Uma-nova-Origem-das-Esp\u00e9cies.jpg&apos;);background-position:76% 90%;background-size:cover;\"><blockquote><p><strong>\u201cEmbora muitos organismos possuam habilidades regenerativas, o envelhecimento \u00e9 um processo biol\u00f3gico praticamente universal que leva a um decl\u00ednio gradual da fun\u00e7\u00e3o fisiol\u00f3gica e ao aumento da vulnerabilidade \u00e0 morte.\u201d<\/strong><\/p><cite>&#8211; (L\u00f3pez et al., 2013) The hallmarks of aging<\/cite><\/blockquote><\/figure>\n\n\n\n<h4 class=\"wp-block-heading has-medium-font-size\"><strong>Senol\u00edticos  e seus desafios<\/strong><\/h4>\n\n\n\n<p class=\"has-black-color has-text-color has-link-color has-medium-font-size wp-elements-ecc1a813c7511bc82b71fd0c6b6c7908\">Com o avan\u00e7o da ci\u00eancia, pesquisadores passaram a investigar formas de eliminar seletivamente essas c\u00e9lulas senescentes. Assim surgiram os chamados senol\u00edticos \u2014 f\u00e1rmacos experimentais que visam reverter efeitos do envelhecimento ao remover essas c\u00e9lulas disfuncionais. Embora promissores, esses compostos ainda enfrentam desafios quanto \u00e0 especificidade e \u00e0 seguran\u00e7a em longo prazo.<\/p>\n\n\n\n<p class=\"has-black-color has-text-color has-link-color has-medium-font-size wp-elements-1db98702c03ac5f2c9819130eb2a9bcf\">Por outro lado, diversos animais nos mostram que os limites da longevidade podem variar de forma not\u00e1vel. Hidras, por exemplo, demonstram aus\u00eancia de senesc\u00eancia detect\u00e1vel. J\u00e1 a tartaruga-gigante das Gal\u00e1pagos pode viver mais de um s\u00e9culo, mantendo fun\u00e7\u00f5es fisiol\u00f3gicas est\u00e1veis por d\u00e9cadas. Embora esses casos inspirem a busca pela extens\u00e3o da vida, eles tamb\u00e9m revelam que a longevidade est\u00e1 profundamente enraizada na gen\u00e9tica e na ecologia de cada esp\u00e9cie (Jones et al., 2014).<\/p>\n\n\n\n<p class=\"has-black-color has-text-color has-link-color has-medium-font-size wp-elements-6dab78b3fa807cf065f3d4736dc6ced5\">Al\u00e9m disso, o envelhecimento humano n\u00e3o se resume \u00e0 biologia celular.
